    Muscarinic cholinergic stimulation of phosphatidylinositol turnover in the longitudinal smooth muscle of guinea-pig ileum

    1. The metabolism of phosphatidylinositol and phosphatidate was investigated in fragments of longitudinal smooth muscle from guinea-pig ileum incubated with cholinergic and anticholinergic drugs. 2. Incorporation of Pi into these lipids was enhanced by acetylcholine and carbamoylcholine. 3. The receptor responsible for triggering this response was of the muscarinic type, since (a) the response was also produced by the muscarinic agonists acetyl-beta-methylcholine, carbamoyl-beta-methylcholine and pilocarpine, and (b) the response was prevented by atropine and prophylbenzilylcholine mustard, but not by tubocurarine. 4. Increased phosphatidylinositol labellin was clearly observed within 5 min in tissue treated with a high concentration of carbamoylcholine. 5. Halfmaximal stimulation of phosphatidylinositol labelling occurred at approx. 10 muM-muM-carbamoylcholine. 6. Incubation of muscle fragments with carbamoylcholine provoked a decrease in phosphatidylinositol concentration, as would be expected if phosphatidyl-inositol breakdown is the reaction controlled by agonists. 7. This information all appears consistent with the proposal that phosphatidylinositol breakdown may be a reaction intrinsic to the mechanisms of muscarinic cholinergic receptor systems

One, the epistemologic aspiration, resting on the search for scientific truth, and two, the social aspiration, resting on the demonstration of a capacity to contribute to psychological assessment problems in particular. Psychometrics answer essentially to psychology's demand for social utility, leading to the quasi-exclusive attribution of importance to quantitative interpretation. For psychology to be considered an empirical science, it has to establish its capacity for the measurement of psychological phenomena, even if this means that it recognizes that these phenomena are essentially qualitative

    Unsteady effects during resistance tests on a ship model in a towing tank

    It is known that there are oscillations in the wave resistance during the constantvelocity phase of a towing-tank resistance test on a ship model. In this work, the unsteady thin-ship resistance theory has been applied to this case. The results have been compared with experiment data obtained using a towing carriage the velocity history of which can be programmed. It is demonstrated here that generally excellent correlation exists between the theory and the experiments. In particular, one can predict the influence of Froude number, rate of acceleration, and type of smoothing of the acceleration on the characteristics of the oscillations. These characteristics include the amplitude, rate of decay, frequency, and phasing of the oscillations in the curve of wave resistance versus time

    How Black Holes Form in High Energy Collisions

    We elucidate how black holes form in trans-Planckian collisions. In the rest frame of one of the incident particles, the gravitational field of the other, which is rapidly moving, looks like a gravitational shock wave. The shock wave focuses the target particle down to a much smaller impact parameter. In turn, the gravitational field of the target particle captures the projectile when the resultant impact parameter is smaller than its own Schwarzschild radius, forming a black hole.     Molecular features of lipoprotein CD0873 - a potential vaccine against the human pathogen Clostridioides difficile

Available on open access from the American Society for Biochemistry and Molecular Biology via the DOI in this recordClostridioides difficile is the primary cause of antibiotic-associated diarrhoea and colitis, a healthcare-associated intestinal disease resulting in a significant fatality rate. Colonization of the gut is critical for C. difficile pathogenesis, and the bacterial molecules essential for efficient colonization therefore offer great potential as vaccine candidates. Here we present findings demonstrating that the C. difficile immunogenic lipoprotein CD0873 plays a critical role in pathogen success in vivo. We found that in a dixenic colonization model, a CD0873-positive strain of C. difficile significantly outcompeted a CD0873-negative strain. Immunization of mice with recombinant CD0873 prevented long-term gut colonization and was correlated with a strong secretory IgA immune response. We further present high-resolution crystal structures of CD0873, at 1.80-2.50 Å resolutions, offering a first view of the ligand-binding pocket of CD0873 and provide evidence that this lipoprotein adhesin is part of a tyrosine import system, an amino acid key in C. difficile infection. These findings suggest that CD0873 could serve as a effective component in a vaccine against C. difficile

    EMOCS: evolutionary multi-objective optimisation for clinical scorecard generation

The final version is available from ACM via the DOI in this recordClinical scorecards of risk factors associated with disease severity or mortality outcome are used by clinicians to make treatment decisions and optimize resources. This study develops an automated tool or framework based on evolutionary algorithms for the derivation of scorecards from clinical data. The techniques employed are based on the NSGA-II Multi-objective Optimization Genetic Algorithm (GA) which optimizes the Pareto-front of two clinically-relevant scorecard objectives, size and accuracy. Three automated methods are presented which improve on previous manually derived scorecards. The first is a hybrid algorithm which uses the GA for feature selection and a decision tree for scorecard generation. In the second, the GA generates the full scorecard. The third is an extended full scoring system in which the GA also generates the scorecard scores. In this system combinations of features and thresholds for each scorecard point are selected by the algorithm and the evolutionary process is used to discover near-optimal Pareto-fronts of scorecards for exploration by expert decision makers. This is shown to produce scorecards that improve upon a human derived example for C.Difficile, an important infection found globally in communities and hospitals, although the methods described are applicable to any disease where the required data is available.Engineering and Physical Sciences Research Council (EPSRC
